LY3437943, a novel triple glucagon, GIP, and GLP-1 receptor agonist for glycemic control and weight loss: From discovery to clinical proof of concept

Coskun T, Urva S, Roell WC, et al. Cell Metabolism 2022;34:1234-1247.e9.

What it found

In cell assays the molecule activated the glucagon and GLP-1 receptors to a similar degree and the GIP receptor more strongly. In obese mice it lowered body weight and blood sugar, and adding glucagon receptor activity increased energy expenditure on top of the lower food intake. In a phase 1 single-dose study in people, its blood levels supported once-weekly dosing and weight stayed lower up to day 43.

Limitations

Mostly cell and mouse data; the human part was a small single-dose safety study. All work by the developer.
