Randomised controlled trial

Retatrutide in adults with obesity and type 2 diabetes (TRIUMPH-2): a double-blind, parallel-group, randomised, placebo-controlled, phase 3 trial

Bellido V, le Roux CW, Ekinci EI, et al. The Lancet 2026;408:1455-1472.

What it found

Mean weight change of -11.9% (4 mg), -16.8% (9 mg) and -18.8% (12 mg) vs -5.1% with placebo. Diarrhoea (27-34% vs 13%) and nausea (14-28% vs 8%) were more common on retatrutide, as were low blood pressure (up to 6% vs under 1%) and dysaesthesia, an abnormal skin sensation (4-7% vs 1%). Stopping because of side effects or death: 12% on 9 mg and 8% on 12 mg vs 5% on placebo.

Limitations

Weight loss was smaller than in people without diabetes. 80 weeks; longer-term safety and outcome data are not yet available. Funded and run by the developer.
