Randomised controlled trial

Semaglutide 2·4 mg once a week in adults with overweight or obesity, and type 2 diabetes (STEP 2): a randomised, double-blind, double-dummy, placebo-controlled, phase 3 trial

Davies M, Færch L, Jeppesen OK, et al. The Lancet 2021;397:971-984.

What it found

Mean body weight change of -9.6% with 2.4 mg vs -3.4% with placebo; 68.8% vs 28.5% lost at least 5%. Gastrointestinal events were reported by 63.5% (2.4 mg), 57.5% (1.0 mg) and 34.3% (placebo), mostly mild to moderate.

Limitations

Weight loss was smaller than in people without diabetes (STEP 1). All groups had lifestyle intervention. Results after stopping were not studied.
