Effect of Continued Weekly Subcutaneous Semaglutide vs Placebo on Weight Loss Maintenance in Adults With Overweight or Obesity: The STEP 4 Randomized Clinical Trial

Rubino D, Abrahamsson N, Davies M, et al. JAMA 2021;325:1414-1425.

What it found

After everyone lost a mean 10.6% in the run-in, those who continued semaglutide lost a further 7.9% while those switched to placebo regained 6.9% (difference 14.8 percentage points). Gastrointestinal events: 49.1% with continued semaglutide vs 26.1% with placebo.

Limitations

Only people who tolerated the full dose in the run-in were randomised, which favours the drug. Adults without diabetes; lifestyle intervention in both groups.
