Randomised controlled trial

Efficacy and safety of once-weekly semaglutide monotherapy versus placebo in patients with type 2 diabetes (SUSTAIN 1): a double-blind, randomised, placebo-controlled, parallel-group, multinational, multicentre phase 3a trial

Sorli C, Harashima SI, Tsoukas GM, et al. The Lancet Diabetes & Endocrinology 2017;5:251-260.

What it found

In adults with type 2 diabetes not yet on glucose-lowering drugs (mean HbA1c 8.05%), HbA1c fell by 1.45% (0.5 mg) and 1.55% (1.0 mg) vs 0.02% with placebo. Body weight fell by 3.73 kg and 4.53 kg on the two doses, more than with placebo. Gastrointestinal events such as nausea were the main reason for stopping treatment.

Limitations

30 weeks only and a small placebo group. Participants were early in their diabetes and on diet and exercise alone, so results may differ in people with longer-standing disease.
